    In situ evidence for renitrification in the Arctic lower stratosphere during the polar aura validation experiment (PAVE)

    In-situ measurements of nitric acid (HNO3), ozone (O3), and nitrous oxide (N2O) were made from the NASA DC-8 during the Polar Aura Validation Experiment in January/February 2005. In the lower stratosphere (9–12.5 km, potential temperature 300–350 K) characteristic compact relationships were observed between all three gases. The ratio HNO3/O3 averaged 3.5 (±0.7) ppt/ppb. Samples with enhanced HNO3/O3 (\u3e4.0) were most abundant under the edge of the Arctic Polar vortex in airmasses with enhanced mixing ratios of both gases (\u3e400 ppb O3 and \u3e2000 ppt HNO3) and reduced mixing ratios of N2O (\u3c305 ppb), indicating air from higher levels in the stratosphere. Relationships to N2O in the anomalous samples under the vortex edge indicate that increases in HNO3/O3 reflect renitrification at DC-8 flight levels, with no indication of significant O3 loss. Renitrified air was only observed at potential temperatures above 340 K, and was most abundant on the PAVE flights on 27 and 29 January

    Impact of multiscale dynamical processes and mixing on the chemical composition of the upper troposphere and lower stratosphere during the Intercontinental Chemical Transport Experiment–North America

    We use high-frequency in situ observations made from the DC8 to examine fine-scale tracer structure and correlations observed in the upper troposphere and lower stratosphere during INTEX-NA. Two flights of the NASA DC-8 are compared and contrasted. Chemical data from the DC-8 flight on 18 July show evidence for interleaving and mixing of polluted and stratospheric air masses in the vicinity of the subtropical jet in the upper troposphere, while on 2 August the DC-8 flew through a polluted upper troposphere and a lowermost stratosphere that showed evidence of an intrusion of polluted air. We compare data from both flights with RAQMS 3-D global meteorological and chemical model fields to establish dynamical context and to diagnose processes regulating the degree of mixing on each day. We also use trajectory mapping of the model fields to show that filamentary structure due to upstream strain deformation contributes to tracer variability observed in the upper troposphere. An Eulerian measure of strain versus rotation in the large-scale flow is found useful in predicting filamentary structure in the vicinity of the jet. Higher-frequency (6–24 km) tracer variability is attributed to buoyancy wave oscillations in the vicinity of the jet, whose turbulent dissipation leads to efficient mixing across tracer gradients

    A Population-Based Surveillance Study of Shared Genotypes of Escherichia coli Isolates from Retail Meat and Suspected Cases of Urinary Tract Infections.

    There is increasing evidence that retail food may serve as a source of Escherichia coli that causes community-acquired urinary tract infections, but the impact of this source in a community is not known. We conducted a prospective, population-based study in one community to examine the frequency of recovery of uropathogenic E. coli genotypes from retail meat samples. We analyzed E. coli isolates from consecutively collected urine samples of patients suspected to have urinary tract infections (UTIs) at a university-affiliated health service and retail meat samples from the same geographic region. We genotyped all E. coli isolates by multilocus sequence typing (MLST) and tested them for antimicrobial susceptibility. From 2016 to 2017, we cultured 233 E. coli isolates from 230 (21%) of 1,087 urine samples and 177 E. coli isolates from 120 (28%) of 427 retail meat samples. Urine samples contained 61 sequence types (STs), and meat samples had 95 STs; 12 STs (ST10, ST38, ST69, ST80, ST88, ST101, ST117, ST131, ST569, ST906, ST1844, and ST2562) were common to both. Thirty-five (81%) of 43 meat isolates among the 12 STs were from poultry. Among 94 isolates in the 12 STs, 26 (60%) of 43 retail meat isolates and 15 (29%) of 51 human isolates were pan-susceptible (P < 0.005). We found that 21% of E. coli isolates from suspected cases of UTIs belonged to STs found in poultry. Poultry may serve as a possible reservoir of uropathogenic E. coli (UPEC). Additional studies are needed to demonstrate transmission pathways of these UPEC genotypes and their food sources.IMPORTANCE Community-acquired urinary tract infection caused by Escherichia coli is one of the most common infectious diseases in the United States, affecting approximately seven million women and costing approximately 11.6 billion dollars annually. In addition, antibiotic resistance among E. coli bacteria causing urinary tract infection continues to increase, which greatly complicates treatment. Identifying sources of uropathogenic E. coli and implementing prevention measures are essential. However, the reservoirs of uropathogenic E. coli have not been well defined. This study demonstrated that poultry sold in retail stores may serve as one possible source of uropathogenic E. coli This finding adds to a growing body of evidence that suggests that urinary tract infection may be a food-borne disease. More research in this area can lead to the development of preventive strategies to control this common and costly infectious disease

    New Record for \u3ci\u3eWoldstedtius flavolineatus\u3c/i\u3e (Ichneumonidae: Diplazontinae), a Hymenopteran Parasitoid of Syrphid Flies in Hawaii

    The parasitoid wasp Woldstedtius flavolineatus (Gravenhorst) (Ichneumonidae) attacks the larvae of syrphid flies (Syrphidae). Woldstedtius flavolineatus was collected in Hawaii for the first time during an extensive malaise trap-based survey of parasitoids in Hawaiian forests. Since its initial collection on Hawaii Island in January 2006, it has been collected at five additional sites on Hawaii Island and at one site each on Maui and Oahu. Malaise trap results from Hakalau Forest National Wildlife Refuge showed a strong seasonal pattern of abundance, with peak population levels reached during July–September. Rearing of its host, Allograpta obliqua (Say), collected from koa (Acacia koa Gray) at Hakalau over two days, revealed a parasitism rate of approximately 95%. Broader impacts of this alien wasp are unknown, but a reduction in host syrphid abundance could result in an increase in numbers of psyllids and aphids (Homoptera) that are preyed upon by syrphid larvae. Furthermore, a reduction in adult syrphids could impact the reproductive success of some of the plants they pollinate

    Inherent biomechanical traits enable infective filariae to disseminate through collecting lymphatic vessels

    Filariases are diseases caused by arthropod-borne filaria nematodes. The related pathologies depend on the location of the infective larvae when their migration, the asymptomatic and least studied phase of the disease, comes to an end. To determine factors assisting in filariae dissemination, we image Litomosoides sigmodontis infective larvae during their escape from the skin. Burrowing through the dermis filariae exclusively enter pre-collecting lymphatics by mechanical disruption of their wall. Once inside collectors, their rapid and unidirectional movement towards the lymph node is supported by the morphology of lymphatic valves. In a microfluidic maze mimicking lymphatic vessels, filariae follow the direction of the flow, the first biomechanical factor capable of helminth guidance within the host. Finally, non-infective nematodes that rely on universal morpho-physiological cues alone also migrate through the dermis, and break in lymphatics, indicating that the ability to spread by the lymphatic route is an ancestral trait rather than acquired parasitic adaptation
